An Animal Model of Extrahepatic Portal Hypertension



Abstract
In preliminary study of this animal experiment by partial portal vein ligation, there was much variation in mortality rate of experimental animal and first it was necessary to fined out reproducibility(or survival) of this animal model. Out of
about 8
week old male Sprague Dawley rats, more than 67% survived after partial ligation of portal vein with the diameter more than 0.8 mm and no death occured after 7 days.
@ES Gross and histologic examination of these rats reveled :
@EN 1) Major collaterals developed around portal vein, on retroperitoneum between spleen and kidney and around esophagus, since at least 7 days after operation.
2) No esophageal varix, portal vein thrombus or ascites developed.
3) The spleen was enlarged between 5~10 days after operation, and after then returned to normal size.
4) Intestine showed ischemic necrosis in rats died within hours.
5) Liver showed mild ischemic hepatitis in rats sacrificed within 3 days.
The acute changes within days suggest that similar acute symptom may occur in acute stage of portal vein thrombosis in infancy or childhood and possible association with necrotizing enterocolitis.
